Gene expression is regulated by epigenetic modifications, which occur either on histones (associated with DNA), on chromatin, or directly on DNA. These modifications include:

  • Methylation of DNA (transcriptional silencing)
  • Histone methylation

  • 5-mC - addition of a methyl group at the 5 position of cytosines is associated with gene silencing
  • 5-hmC - is a DNA modification and results from the enzymatic oxidation of 5-mC by the TET family of cytosine oxygenases. 5-hmC modification at a gene is associated with its expression level
