Application example
10 or 20 µg of soluble protein extract from WT-Col-0 Arabidopsis thaliana extracted in a buffer containing 50 mM HEPES, 5 mM NaCl and 10 mM MgCl2, separated on 12% SDS-PAGE and blotted 1h to PVDF using semi-dry transfer. Blots were blocked with 4% milk for 1h at room temperature (RT) with agitation. Blot was incubated in the primary antibody at a dilution of 1: 1 000 overnight in 4°C with agitation. The antibody solution was decanted and the blot was 3 times for 5 min in TBS-T at RT with agitation. Blot was incubated in secondary antibody (anti-rabbit IgG horse radish peroxidase conjugated, from Agrisera AS09 602) diluted to 1:20 000 in for 2h at RT with agitation. The blot was washed as above and developed for 5 min with ECL according to the manufacturer's instructions. Exposure time was 10 min.

Additional information (application): 5 mM DTT in extraction buffer and 5% B-ME in Lämmli buffer are recommended to use, Samples should be heated at 95°C for 2 min before loading as TRXs proteins have a tendency to oligomerize - Background

Bettina Mathes | 2016-06-23With a 1:1000 dilution, 4% Milk in 1xTBS with 0.05% Tween (30 minutes blocking) the antibody works fine on pea stroma and chloroplasts and with Arabidopsis chloroplasts. It's only important to cook the samples before for 3 minutes at 95°C. Incubation 2h at roomtemperature is enough.
