Application example
Material: Barley(flag leaves), fixed in 4% formaldehyde (from paraformaldehyde), embedded in LR White
Samples: Ultrathin sections on Au-Grids
Immunogold labelling:
1. Blocking 30 min RT in PBS (pH 7,4) + 0,2% BSA + 0,2 % fish gelatin (= blocking buffer) sterile filtrated
2. Incubation over night at 4°C anti-W1 (goat) 1:200 in blocking buffer
3. Washing: 4x 3 min PBS + 0,1 % BSA (= washing buffer)
4. Incubation 60 min RT, dilution: 1:50 in blocking buffer, rabbit anti-goat (10 nm gold) von Aurion (Code 810077)
5. Washing: 3x 5 min PBS
6. Washing 4x2 min mit A. millipore, staining in saturated aqueous uranyl acetate 10 mins

Background: WHIRLY proteins are multifunctional DNA/RNA-binding proteins (Prikryl et al. 2008, NAR 36: 5152-5165). WHIRLY1 was shown to be located both in chloroplasts and nucleus (Grabowski et al. 2008, Plant Physiology 147:1800-1804). In chloroplasts it is part of nucleoids (Pfalz et al. 2006, Plant Cell 18:176-197; Melonek et al. 2010, Planta 232:471-481). In the nucleus, WHIRLY1 is an acitivator of pathogen response genes (Desveaux et al. 2000, Plant Cell 12:1477-1489). WHIRLY2 is a mitochondrial protein and affects DNA copy number (Cai et al. 2015, Plant Physiology 169:660-673). - Product Citations
