application example
Chlamydomonas reinhardtii soluble proteins (10 µg) were separated on a 7.5% SDS-PAGE gel and blotted to nitrocellulose for 90 min. at 1.5 mA cm-2. The membrane was blocked with 1% milk in PBS-T for 1h at room temperature (RT) with agitation. Blot was incubated in the primary antibody at a dilution of 1: 1 000 for 2 hr at RT with agitation. The antibody solution was
decanted and the blot was rinsed briefly twice, then washed 3 times for 5 min in PBS-T + 1% milk at RT with agitation. Blot was incubated in secondary antibody (anti-rabbit IgG horse alkaline phosphatase conjugated, from Southern Biotech ) diluted to 1:3000 in PBS-T + 1% milk for 45 min at RT with agitation. The membrane was washed 2 times for 5 min in PBS-T + 1% milk at RT with agitation, then rinsed with TBS (pH 7.5), and developed.Courtesy Dr. Dudley Page, UCLA, USA
