This guide aims to provide you with more information on different tags, as well as tag antibody recommendations.
Common tags
• Affinity Tag (AT): Binds specifically to a partner molecule, allowing one-step purification of the protein using affinity chromatography methods.
• Fluorescent Tag (FT): Enables the detection and localization of proteins through their ability to emit fluorescence when excited by specific wavelengths of light.
• Fuctional/Enzyme Tag (FET): Has a biological activity, such as enzymatic or catalytic properties, often used for performing specific biochemical reactions or assays.
• Reporter Protein Tag (RPT): Provides a measurable signal, allowing monitoring of the expression or activity of a protein in a system.
• Selectable Marker Tag (SMT): Enables selection or differentiation based on resistance to a specific substance.
• Solubility/Expression Enhancer Tag (SEET): Improves the solubility, stability, or expression of the target protein in systems where proteins may otherwise aggregate or be poorly expressed.

Tag antibodies Q&A
Most tag antibodies are species-independent since they recognize artificial peptide sequences, not endogenous proteins.
Tag antibodies bind specifically to the tagged protein. These antibody-protein complexes can be pulled down using beads, isolating the tagged protein and its interacting partners.
Antibodies can be utilized in live cell imaging, using flow cytometry, to detect membrane surface proteins. In this application, fluorescent tags like GFP are commonly used. Antibody-based detection typically requires cell fixation and permeabilization, making it unsuitable for live cells unless specialized cell-permeable antibodies are used.
Proximity labelling is a laboratory technique which makes it possible to label proteins (in this case by biotinylation), proximal to protein of interest. This allows the study of protein interactome networks in different types of cells and organs, as well as different developmental stages. The technique is achieved by a gene fusion in a living cell, between the protein of interest and BioID (BirA). The TurboID variant is a biotin ligase, with substantially higher activity, which provides higher temporal resolution and broader application in vivo. The labelling time is just 10 minutes in RT, compared to the over 18 hours of labelling time required for BioID.
